СОВРЕМЕННЫЕ ТЕНДЕНЦИИ В РАЗРАБОТКЕ И ПРОИЗВОДСТВЕ НАНОРАЗМЕРНЫХ СИСТЕМ ДЛЯ ДОСТАВКИ ЛЕКАРСТВЕННЫХ СОЕДИНЕНИЙ

Аннотация
Разработка систем доставки лекарственных средств является одним из приоритетных направлений развития фармацевтических технологий. В настоящее время основная часть крупнейших фармацевтических компаний работает в области разработки систем доставки как для новых лекарственных препаратов, так и для уже имеющихся на рынке. Одной из ведущих областей их применения была и по-прежнему остается онкология. Разнообразие задач, решаемых при разработке систем доставки, и способов их решения привело к появлению многочисленных вариантов таких систем, каждая из которых имеет свои плюсы и минусы. Специфичность систем доставки и их разнообразие привело к необходимости систематизации сведений как об их физико-технологических характеристиках, так и о возможностях их применения в клинической практике. В обзоре обобщаются и систематизируются сведения о современных адресных системах доставки для лекарственных соединений с низкой биодоступностью, описанных в научной литературе и применяемых в современной фармацевтической технологии. Показаны тенденции в разработке наноразмерных систем доставки для различных терапевтических препаратов, способных к проникновению через защитные барьеры организма, к устойчивому контролируемому высвобождению, а также перспективных для целевой доставки в клетки-мишени. Наноразмерные системы доставки лекарственных средств обладают большим потенциалом для фармацевтической и медицинской промышленности. Выделены следующие наноразмерные системы для доставки лекарственных препаратов: наноэмульсии, нанокапсулы, нанолипосомы (в том числе, экзосомы, виросомы и прочие модификации традиционных липосом), дендримеры, а также носители на основе клеток и пептидов. Приведены их основные технологические и фармакологические характеристики, рассмотрены перспективы применения в клинической практике. Приведено описание основных методик формирования для каждой из представленных систем доставки. Показано, что ежегодно увеличивается интерес к модификациям липосомальных систем, в частности к экзосомам, а также к дендримерам и системам доставки на основе клеток.
